It is advisable not to take backup while you are using the computer. The backup might fail if it tried to backup the files which are currently in use.
Choose a time when you wont be using the computer. My computer runs the backup automatically at 11 PM every night.
Close all the files, photos and software including your email client software such as Outlook, Windows Live Mail, etc.
